Protein Properties: Homo- and Heterogeneity

Homogeneity is one of the most basic and important protein properties in general life science and biopharmaceutical applications. In an ideal study, the protein of interest would exist throughout the sample in a consistent form and size to ensure optimal purity for accurate characterization. This enables chemists to precisely identify protein properties for specific downstream applications. Yet proteins are unlikely to exist in this ideal state for long, given the sheer variety of potential aggregating and post-translational modification phenomena that can affect the sample.

Protein heterogeneity is a complex property characterized by both transient and permanent forms of modification – which are also subject to individual variations throughout a distinct protein’s lifespan. The resultant range of heterogeneous variability is subsequently vast. It extends far beyond the potential for modification suggested by the underlying amino acids in the polymeric chain alone.

Analyzing these essential protein properties is both challenging and essential. Homo- and heterogeneity of protein samples may be indicative of purity, long-term stability, informational content, physical and mechanical behavior in formulation, and more.
