Is your compound extremely polar? Use HILIC!
HILIC is one of the most successful techniques for the separation and quantitation of polar organic compounds. It is most commonly applied for molecules which are too polar to be retained by reverse phase chromatography. In HILIC, the stationary phase is strongly polar (typically ionic, polyamide or silica) and uses a high percentage of organic solvent (acetonitrile). This results in formation of an enriched water layer around the stationary phase. Partitioning between the water layer and the bulk mobile phase leads to retention and causes separation of sample components.
- Retention of hydrophilic compounds that are difficult to retain by Reversed Phase Liquid Chromatography
- Efficient separation of very polar analytes
