Isolation, Purification and Structure Confirmation. Fraction collection is typically performed for the purpose of chemical structure confirmation or to collect enough of a chemical to serve as a reference standard. Fraction collection allows a single peak in a chromatogram to be isolated. Nearly all of the modes of chromatography are amenable to fraction collection including Size Exclusion Chromatography (SEC), Reverse Phase (RP), normal phase (NP) and Hydrophilic interaction chromatography (HILIC). Methods which use salts or non-volatile buffers generally are not preferred. Fraction collection is typically appropriate to obtain nanogram to gram quantities of an analyte depending on sample viscosity and concentration of the desired analyte in the starting sample solution.
- Isolation of desired chemical species on the nanogram to gram scale
- Structure confirmation
- Purification of reference standards
